The process affected by chloroquine was estimated to have a T1/2 of 8 h. Cells grown in the presence of cardiac glycosides accumulate large numbers of glycoside molecules; chloroquine, NH4Cl and amantadine increase the accumulation of digoxin and digitoxin and may decrease that of ouabain. Chloroquine is used to prevent or treat malaria caused by mosquito bites in countries where malaria is common. Malaria parasites can enter the body through these mosquito bites, and then live in body tissues such as red blood cells or the liver. The usefulness of hypokalemia as an indicator in the evaluation of chloroquine toxicity was studied in a retrospective series of 191 acute chloroquine poisonings. Results indicated that the risk of severe poisoning and death are proportional to the degree of hypokalemia.

Lack of awareness and recognition of clinically significant DDIs is a major obstacle to safe ARV prescribing. This review undertakes a systematic evaluation of potential DDIs between ARVs and drugs used to treat TB, malaria, chronic Hepatitis B&C infections and opioid dependence. Before the administration of chloroquine, the patient had only a mild skin erythema in the irradiated area, which was consistent with the radiotherapy dose she had received. On day 3 of chloroquine therapy, she developed localized brisk bullous eruptions in the irradiated area, which developed into a patch of fulminant moist desquamation. Chloroquine may increase digoxin levels; increased digoxin monitoring is warranted. Use of chloroquine could possibly also lead to increased levels of calcineurin inhibitors and should be used with caution.
