Oct 01, 2018 Chloroquine phosphate tablets, Chloroquine phosphate, USP, is a 4-aminoquinoline compound for oral administration. It is a white, odorless, bitter tasting, crystalline substance, freely soluble inwater. Chloroquine phosphate tablets are an antimalarial and amebicidal drug. Uses Chloroquine is used to prevent or treat malaria caused by mosquito bites in countries where malaria is common. Malaria parasites can enter the body through these mosquito bites, and then live. The clinical usefulness of chloroquine, and in some recent cases of quinine as well, has been much reduced by the evolution and spread of chloroquine resistant malaria parasites. The mechanism of resistance involves a reduced accumulation of the drug, although again the mechanism involved is controversial.

Bafilomycin A1 and chloroquine are commonly used compounds that inhibit autophagy by targeting the lysosomes but through distinct mechanisms. Pharmacological modulators of this pathway have been extensively utilized in a wide range of basic research and pre-clinical studies.

Chloroquine is a lysosomotropic agent as it is usually prepared as a diprotic weak base pKa 8.5. The unprotonated form of chloroquine preferentially accumulates in lysosomes as it rapidly diffuses across cell/organelle membranes. Chloroquine is also a lysosomotropic agent, meaning it accumulates preferentially in the lysosomes of cells in the body. The pK a for the quinoline nitrogen of chloroquine is 8.5, meaning it is about 10% deprotonated at physiological pH as calculated by the Henderson-Hasselbalch equation. Chloroquine is used to treat and to prevent malaria. Chloroquine is also used to treat amebiasis infection caused by amoebae. Chloroquine may also be used for purposes not listed in this medication guide.
